基于神经网络模型评价淫羊藿、女贞子配伍对自然衰老大鼠肾脏、睾丸及肾上腺的影响

摘要:

目的:研究自然衰老大鼠肾脏、睾丸和肾上腺的病理变化、肾脏自由基损伤的增龄性改变,以及中药淫羊藿、女贞子对其的影响,基于神经网络模型评价药物作用的差异。方法:8月龄雄性SD大鼠,随机分为7组,包括9月龄组、12月龄组、15月龄组、17月龄组、淫羊藿组、女贞子组、淫羊藿女贞子组(配伍组);另以3月、6月龄雄性大鼠为青年对照组。3个用药组饲养至15月龄后开始灌胃给予相应药物2个月后处死,其他组大鼠饲养至相应月龄后处死。剖取肾脏、肾上腺及睾丸,HE染色观察病理改变;制备肾脏匀浆,检测相关指标与水平的变化。结果:与青年大鼠比较,17月龄组老年大鼠肾脏、睾丸及肾上腺出现退行性的病理改变,且肾脏组织一氧化氮(NO)、总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)、还原型谷胱甘肽(GSH)水平降低,总一氧化氮合成酶(tNOS)、诱导型一氧化氮合成酶(i NOS)及丙二醛(MDA)水平升高(P<0.05或P<0.01)。与17月龄组比较,配伍组肾上腺及睾丸退行性病理改变减轻,肾脏组织T-AOC、SOD、GSH、NO水平升高,tNOS、iNOS及MDA水平降低(P<0.05或P<0.01)。基于多层感知(MLP)神经网络模型评价药物的综合作用,结果显示,淫羊藿、女贞子配伍可使17月龄自然衰老大鼠的上述指标改变逆转至(9.79±1.44)个月。结论:淫羊藿、女贞子配伍具有延缓衰老的作用,可能与改善衰老大鼠肾上腺及睾丸退行性病理改变、提高肾脏抗氧化能力有关。

Abstract:

Objective:To study the patholigical changes of the pathology of kidney,testicle and adrenal gland and the free radical damage of kidney in natural aging rats,and the effect of Yinyanghuo(Herba Epimedii) and Nvzhenzi(Fructus Ligustrilucidi) on the changes,and to evaluate the therapeutic effect of drugs based on the neural network model.Methods:The 8-month male SD rats were randomly divided into 7 groups,including 9-month group,12-month group,15-month group,17-month group,Yinyanghuo group,Nvzhenzi group and combination group.The 3-month and6-month male rats were used as the youth control group.Rats in the three treatment groups were administered with corresponding drugs respectively by gavage for two months after being fed to 15 months old,and rats in the other groups were fed to corresponding months old.And then,all rats were sacrificed and their kidneys,adrenal gland and testis were taken for observing pathology changes by HE staining.The levels of related indicators such as nitric oxide(NO),total nitric oxide synthase(tNOS),inducible nitric oxide synthase(iNOS),constructive nitric oxide synthase(cNOS),total antioxidant capacity(T-AOC),superoxide dismutase(SOD),malondialdehyde(MDA) and glutathione(GSH) in the rats' kidneys were detected.Results:Compared with the young rats,degenerative pathological changes of kidney,testicle and adrenal gland were shown,the levels of NO,SOD,GSH and T-AOC were decreased significantly(P<0.05 or 0.01) and the levels of tNOS,iNOS and MDA were increased significantly(P<0.05 or 0.01) in 17-month group.Compared with 17-month group,the combination group could improve degenerative pathological changes of adrenal gland and testis,increase the levels of SOD,GSH,T-AOC and NO,and decrease the levels of tNOS,iNOS and MDA in kidney(P<0.05 or 0.01).The study based on multi-layer neural network(MLP) model suggested that the combination of Yinyanghuo and Nvzhenzi could reverse the above-mentioned changes of 17-month natural aging rats to(9.79±1.44) months old.Conclusion:The anti-aging effect of the combination of Yinyanghuo and Nvzhenzi may be related to delaying the degeneration of adrenal gland and testis and improving the antioxidant capacity of kidney in aging rats.
